The fermentation conditions for maximum flocculation/settling performance of a BIOTECH flocculent yeast strain, Saccharomyces cerevisiae TB3 was explored in reconstituted sugarcane syrup medium. The factors studied were pH, Ca²⁺ supplementation and NH₄(SO₄)₂ concentration. The responses were % flocculation,settling time and fermentation parameters of cell optical density (OD), total residual sugar (TRS) concentration and ethanol concentration. Also, the ethanol fermentation ability of the strain TB3 at optimum conditions (for maximum flocculation) was compared with the industrial, non-flocculent strain, S. cerevisiae HBY3. The results of the 2ᵏ factorial experiment showed that all the factors were significant. RSM experimental results showed that for all the responses, the recommended statistical models were quadratic. The obtained optimum fermentation conditions were pH 5.5, 0.03% v/v NH₄(SO₄)₂ concentration and no Ca²⁺ supplementation. Under these conditions, the experimental response values (which agreed with the predicted values) were 97.44 % flocculation, 72.50 seconds settling time, 14.30 cell OD, 101.88 mg/mL TRS concentration and 4.57% ethanol concentration. It was also found that S. cerevisiae TB3 exhibited statistically higher ethanol fermentation performance than S. cerevisiae HBY3.
